William F. Anderson is an American physician, geneticist, and molecular biologist recognized as a pioneer of gene therapy. Trained at Harvard and with a stint in Cambridge in Francis Crick’s laboratory, Anderson made early contributions to molecular biology, taking part in deciphering the genetic code under Marshall Nirenberg’s supervision and publishing extensively throughout his career. In the 1970s he developed foundational methods for studying protein synthesis in eukaryotes, isolating initiation factors and creating cell-free systems that made it possible to synthesize human proteins from RNA extracted from reticulocytes. That work paved the way for investigations into genetic diseases such as sickle cell anemia and thalassemia. From the 1980s onward he concentrated on clinical applications of these discoveries, proposing and developing efficient retroviral vectors to deliver genes to hematopoietic cells. In 1990 he coordinated the first successful case of gene therapy in a child with severe combined immunodeficiency, a milestone that solidified his prominence. Author of hundreds of articles and books and recipient of numerous honors, his legacy lies in moving basic molecular biology toward therapeutic interventions that laid the foundations of modern gene therapy.
